Test kit system for diagnostics of brucellosis of animals in the milk ring test (MRT)
Purpose:
Serological diagnosis of animal brucellosis in the milk ring test (MRT).
Description and characteristic:
The kit system components are:
- antigen is a nontransparent liquid of a blue or cherry-red colour with the sediment of microbial cells formed at storage, homogenized at shaking;
- dry brucellosis serum looks like a lyophilized mass of a light-gray colour with a yellowish tone, dissolved in distilled water or normal saline solution within 3 min with a formation of transparent liquid of a light-yellow colour.
This kit includes the following components:
- brucellosis antigen for the MRT. It is a suspended matter of heat-inactivated and phenol-inactivated brucellas of species abortus in 0.85% solution of sodium chloride with 0.5% phenol, haematoxylin-stained in blue or tetrazolium-stained in cherry-red;
- dry brucellosis serum is a blood serum of bulls (oxen) hyperimmunized by a live culture of brucellas of species abortus.
The test-system is used for testing 500 cow milk samples.
Method principle:
The S-brucellosis antigen for the milk ring test is agglutinated by homologous antibodies of milk of cows that are diseased with brucellosis or immunized with brucellosis agglutinogenic vaccines resulting in phenomenon of adsorption of the antigen-antibody complex by milk fat globules and formation of the coloured ring in the cream layer.
The brucellosis serum contains specific agglutinins. It is used as a positive control for the milk ring test.
Production form:
Antigen is packaged into 10 cm3 and sera - in 2 cm3 bottles of suitable capacity.
Storage and transportation conditions:
The test-system should be stored and transported in a dry dark place at a temperature of 2 to 8°C. Transportation of the test-system is admissible at a temperature not above 20°C for no longer than 7 days.
