Test kit system for diagnostics of brucellosis of animals in the Rose Bengal Test (RBT)
Purpose:
Serological diagnosis of animal brucellosis in rose Bengal test.
Description and characteristic:
The kit system components are:
- antigen is a nontransparent liquid of a rose colour with the sediment of microbial cells formed at storage, homogenized at shaking;
- dry brucellosis and negative sera look like a lyophilized mass of a beige colour, dissolved in distilled water or physiological solution within 3 min with a formation of transparent liquid of a light-yellow color.
The kit includes the following components:
- brucellosis antigen for the RBT. It is a suspended matter of inactivated brucellas of species abortus in lactic buffer solution pH 3.65 with 0.5% phenol, Rose Bengal-stained;
- dry brucellosis serum is a blood serum of bulls (oxen) hyperimmunized by a live culture of brucellas of species abortus;
- dry negative blood serum of cattle.
The test-system is used for testing 2000 blood serum samples of cattle, horses, camels or 4000 blood serum samples of small cattle, reindeer (marals) in RBT.
Method principle:
The S-brucellosis antigen for the RBT is agglutinated by homologous antibodies of animal blood serum that are diseased with brucellosis or immunized with brucellosis agglutinogenic vaccines. The brucellosis serum contains specific antibodies agglutinating the antigen in the Rose Bengal test. It is used as a positive control for the RBT. The negative serum does not contain specific antibodies to the brucellosis antigen for the RBT and is used as a negative control for the RBT.
Production form:
Antigen is packaged into 10 cm3 and sera - in 2 cm3 bottles of suitable capacity.
Storage and transportation conditions:
The test-system should be stored and transported in a dry dark place at a temperature of 2 to 8°C. Transportation of the test-system is admissible at a temperature not above 20°C for no longer than 7 days.
Shelf life:
24 months.
