Test system for detecting Influenza A virus RNA subtypes H5/H7/H9 in biological material using reverse transcription and polymerase chain reaction with real-time fluorescence detection.
Purpose
Typing (identification of subtypes H5, H7, H9) of Influenza A viruses (Influenza virus A) in influenza virus cultures in biological material using a combined reverse transcription and RNA amplification method with real-time fluorescence detection in the presence of an internal positive control. The test system is not intended for primary screening for Influenza type A. The test system is designed for the analysis of 55 samples.

Principle of the method
Typing (identification) of subtypes H5, H7, H9 is performed by amplifying regions of the Influenza A virus gene for these subtypes in the presence of three fluorescently-labeled oligonucleotide probes. Fluorescent signal reading is carried out in three detection channels of the instruments directly during PCR. Amplification products are detected in real-time mode using the principle of cleavage of the fluorescent label at the 5' end of the oligonucleotide probe.
Analytical sensitivity of the kit is no less than 10 genome equivalents per reaction (103,3 GE/cm3).
Production form
Components included in the test system are packaged in polypropylene microtubes with screw-on caps with a capacity of 2 cm3 and 0.5 cm3.
Storage and transportation conditions
The test system is transported and stored in the manufacturer's packaging in a light-protected place at a temperature of -18 to 24ºC. Multiple freeze-thaw cycles and transportation at a temperature of 2 to 8°C for up to seven days are permitted.
